The Effects of 3'-deoxyadenosine on the Synthesis of Ribonucleic Acid.

Cordycepin was first isolated from the culture filtrate of the mold Cordyceps militaris by Cunningham et al. (1) and subsequently characterized by Bentley, Cunningham and Spring (2) as 3’-deoxyadenosine with a branched chain. Kaczka et al. (3) have recently isolated from the filtrate of Aspergillus nidulans a material inhibiting the growth of KB cells and identified the active component as 3’-deoxyadenosine with a straight chain 3-deoxyribose moiety. The identity of this substance with synthetic 3’-deoxyadenosine obtained from Dr. Leon Goodman (Stanford Research Institute) and samples of cordycepin supplied by Professor R. A. Raphael (University of Glasgow) and Dr. Chester Stock (Sloan-Kettering Institute) has been proven (4). The effect of 3’-deoxyadenosine on the biosynthesis of ribonucleic acids in intact Ehrlich ascites tumor cells and the effects of 3’deoxyadenosine 5’-mono-, 5’-di-, and 5’-triphosphate on partially purified RNA polymerase from Micrococcus lysodeikticus are described in this paper.
